肠激酶(Enterokinase,light chain)是一种高度专一性识别Asp-Asp-Asp-Asp-Lys序列的蛋白酶,在Lys的C端水解多肽。它可以将胰蛋白酶原转变为胰酶,也可以将带有这个识别序列的融合蛋白切开。由于肠激酶具有高度专一性,水解效率高的特点,它被广泛地应用于基因工程产品的开发,其应用之一是作为工具蛋白酶用于重组融合蛋白质的特异性断裂,尤其适用于生物工程制药业及基因工程、生物化学、分子生物学等研究。本产品为采用重组酵母分泌表达的高纯度、高活性的牛肠激酶,适用范围广(4~45℃,pH4.5~9.5),并且在各种去垢剂和变性剂存在的条件下仍具有部分活性。

常见影响肠激酶活性的因素:

>2M Urea,>250mM NaCl,>20mM β-ME,>0.1% SDS,>50mM imidazole。

如果样品溶液中含有上述成分的一种或多种,为获得理想的酶切结果,请先将样品透析到1×反应缓冲液中,然后再进行酶切实验。

应用举例:
肠激酶EK蛋白消化(Enterokinase)
图例一)重组牛肠激酶酶切反应:每个反应含有15μg的Thioredoxin-NP-27和不同数量的肠激酶。在37°C经过16小时的反应后用15°C的SDS-PAGE胶检测酶切效果。
每个反应所含的酶量分别为:
Lane A: No rEK
Lane B: 0.0008 units
Lane C: 0.0015 units
Lane D: 0.003 units
Lane E: 0.008 units
Lane F: 0.015 units
Lane G: 0.03 units
质量保证:
经多次柱纯化,SDS-PAGE胶检测仅可见清晰单一的目的条带;本制品中不含有非特异性的蛋白酶 切活性。
活性定义:
在37℃,16小时内将0.5毫克的Thioredoxin-NP-27,95%降解为NP-27所需要的酶量定义为1个活性单位。
保存条件:
-20°C可保存一年以上。
1×重组牛肠激酶反应缓冲液:
50mM Tris-HCl,pH 8.0(22℃),1mM CaCl,0.1% Tween-20。
