Phase 1. Large-scale fermentation
Transformation of a customer-supplied vector into high-efficiency expression bacterial strain.
Selection of bacterial colonies for fermentation.
20 liters of high density bacterial culture (about 60g cell pellets per liter culture1) will be induced with IPTG or heat and the cell pellets will be harvested by centrifugation.
Test for the expression of the recombinant protein by SDS-PAGE.
Time: 2 weeks
Phase 2. Cell disruption and inclusion bodies purification
Cell pellets disruption by mechanic homogenate.
Washing the inclusion bodies with standard washing buffer (or customer required buffer).
Test for the washing effect by SDS-PAGE.
Time: 1 week
Phase 3. Protein purification
Protein purification as pilot scale procedure.
Provide from a few grams up to a few kilograms purified protein.
Refolding and endotoxin removing of target protein.
